Development of new male contraceptive methods would be a valuable addition to the limited choices currently available to men, namely condoms, withdrawal, abstinence, and vasectomy. The Council's research on male reproduction and its male contraceptive R&D are unique because they examine men's reproductive health as a whole, rather than focusing exclusively on contraception.
Population Council focus
At the Population Council's Center for Biomedical Research in New York City, male reproductive health projects include:
- Developing subdermal contraceptive implants delivering the novel androgen MENT®, a synthetic steroid that resembles testosterone but does not stimulate prostate growth like testosterone can when used as a contraceptive. Council researchers are using MENT® acetate in contraceptive implants because it suppresses sperm cell development without reducing libido.
- Determining the additional potential of MENT® as a hormone therapy method (via transdermal gel or patch) to treat age-related symptoms associated with the natural decline of testosterone production in men.
- Investigating the biology and regulation of events that occur during Sertoli–Sertoli and Sertoli–germ cell communication during spermatogenesis, with the goal of developing safe, reversible male contraceptives. Currently, adjudin (1-[2,4-dichlorobenzyl]-1H-indazole-3-carbohydrazide) is the lead compound under investigation. Probing studies in male contraception revealed adjudin is capable of depleting germ cells by disrupting cell adhesion in the testis, thereby creating reversible contraception.
- Conducting translational studies to understand the underlying causes of male infertility associated with the arrest of sperm cell development and the absence of sperm (azoospermia). Identifying novel DNA binding proteins, which are critical to successful division in developing germ cells, may provide new lead compounds for therapeutic interventions and provide avenues for a new generation of small molecules for male contraception.
Population Council impact
The development of safe, effective male contraceptive technologies is a new frontier in reproductive health technology with exciting implications—particularly because of their potential to engage men more centrally in the pursuit of reproductive health and gender equity. When Council R&D is complete, these products will be valuable additions to the array of available contraceptive options. In addition:
- Such dedicated research into various formulations of MENT® has resulted in considerable progression of male subdermal implant technology.
- Basic and translational research has identified novel mechanisms of germ cell development in men and has led to greater understanding of DNA and chromosome modifications during human sperm production.
Future directions
- In the search for innovative methods of male contraception, the Council is exploring a novel approach using a molecule that prevents maturation of the sperm, making it unable to fertilize an egg.
- Researchers also are investigating the mechanism by which environmental toxicants—such as cadmium, xenostrogens like bisphenol A (BPA), and phthalates—disrupt spermatogenesis. (Some environmental toxicants can lower sperm count, resulting in male subfertility or infertility.)
